SR-14968 (CAS No. 2133455-40-8) is a synthetic benzimidazolinone u-opioid receptor agonist belonging to the class of experimental “SR-series” orphine-like compounds. It features a halogenated benzimidazol-2-one core with a substituted piperidine side chain and was developed as a tool compound to study ?-opioid receptor signaling, biased agonism, and analgesia–respiratory separation in preclinical models.
Functionally, SR-14968 acts as a high-efficacy ?-opioid receptor agonist. Early in vitro work characterized it as G-protein–biased with relatively weak ?-arrestin-2 recruitment compared to some classical opioids, while subsequent in vivo studies indicate a profile closer to an “unbiased” full agonist with robust antinociceptive efficacy and a somewhat wider separation between analgesic and respiratory-depressant doses in rodents. At sufficiently high exposures, however, SR-14968 still produces typical opioid-like adverse effects, including respiratory suppression, dependence, and withdrawal signs in animal models.
